Membrane dome-based regulation mechanism of the mechanosensitive PIEZO ...

Our findings provide electrophysiological evidence of the membrane dome mechanism by showing that a PIEZO channel with a larger in-plane projection area change is more sensitive to mechanical force, with a nice fit of current-pressure curves between experimental measurement and model simulation.

It can be directly activated by membrane tension, with the peripheral blade and beam structures likely acting as mechanotransduction modules. Notably, PIEZO1 requires lower tension for activation compared to bacterial mechanosensitive channels. The protein exhibits voltage-dependent inactivation.

Here, we show that the mechanosensitive ion channel Piezo1 is essential for the maturation of intercellular junctions into functional continuous adhesions. Deletion of Piezo1 results in an imbalance of cell contractility and membrane tension, leading to a delay in adhesion maturation.

Through membrane mechanical calculations, we show that Piezo1 deforms the membrane shape outside the perimeter of the channel into a curved ‘membrane footprint’. This membrane footprint amplifies the sensitivity of Piezo1 to changes in membrane tension, rendering it exquisitely responsive.

Piezos are large transmembrane proteins conserved among various species, but they don't have known sequence similarity to any other class of proteins. Piezo proteins contain around 2500-2800 amino acids depending on the species. [8]
